Genetic evidence for a species complex within the piranha Serrasalmus maculatus (Characiformes, Serrasalmidae) from three Neotropical river basins based on mitochondrial DNA sequences Genet. Mol. Biol.
Bignotto,Thaís Souto; Maniglia,Thiago Cintra; Gomes,Vivian Nunes; Oliveira,Isadora Janolio de; Agostinho,Carlos Sérgio; Prioli,Sônia Maria Alves Pinto; Prioli,Alberto José.
Abstract Mitochondrial molecular markers (DNA sequences of D-loop, cytochrome b and cytochrome c oxidase I) were employed to characterize populations of the piranha Serrasalmus maculatus from Upper Paraná, Upper Paraguay and Tocantins River basins. D-loop sequences of S. maculatus population from Paraná-Paraguay River basin exhibited tandem repeats of short motifs (12 base pairs) and variable numbers depending on specimens, accounting for length variation. Concatenated mitochondrial sequences suggested that S. maculatus encompasses different mitochondrial DNA lineages. Although sampling was restricted to three river basins, phylogenetic analysis clearly indicated that the species currently recognized as S. maculatus presents high genetic variability....

Genetic structuring of Salminus hilarii Valenciennes, 1850 (Teleostei: Characiformes) in the rio Paraná basin as revealed by microsatellite and mitochondrial DNA markers Neotropical Ichthyology
Silva,Juliana Viana da; Hallerman,Eric M.; Orfão,Laura Helena; Hilsdorf,Alexandre Wagner Silva.
Genetic variation of Salminus hilarii was assessed by screening microsatellite loci and mitochondrial D-loop DNA across four sampling in the upper rio Paraná basin of Brazil. Genetic diversity - measured as mean expected heterozygosity (0.904) and mean number of alleles across populations (13.7) - was reasonably high. Differentiation of microsatellite allele frequencies among populations was shown to be low but significant by AMOVA Φ ST (0.0192), and high by D EST (0.185). D-loop variation was high, with haplotypic diversity of 0.950 and nucleotide diversity of 0.011. Mitochondrial DNA-based estimates for population differentiation were high, with an overall Φ ST of 0.173. The results of tests of nuclear and mitochondrial variation yielded no unequivocal...

Identification of Astyanax altiparanae (Teleostei, Characidae) in the Iguaçu River, Brazil, based on mitochondrial DNA and RAPD markers Genet. Mol. Biol.
Prioli,Sônia M.A.P.; Prioli,Alberto J.; Júlio Jr.,Horácio F.; Pavanelli,Carla S.; Oliveira,Alessandra V. de; Carrer,Helaine; Carraro,Dirce M.; Prioli,Laudenir M..
Astyanax fishes are among the most important food-web components of South America rivers. In the Iguaçu River basin, the Astyanax genus is represented mainly by endemic species. For millions of years, that hydrographic basin has been geographically isolated from the Paraná River basin by the Iguaçu Falls. Recently, a species from the Upper Paraná River basin identified as Astyanax bimaculatus was revised and described as a new species named Astyanax altiparanae Garutti & Britski, 2000. Fauna endemism and geographic isolation triggered interest in investigations to evaluate the identification and genetic relatedness among two A. altiparanae populations from the Upper Paraná River basin and the population identified as A. bimaculatus in the Iguaçu...

Invasive north american Beaver (Castor canadensis): the distribution of mitochondrial variation across the archipelago of Tierra del Fuego Mastozool. neotrop.
Fasanella,Mariana; Poljak,Sebastián; Lizarralde,Marta S..
In 1946 twenty-five pairs of Castor canadensis were introduced into the Isla Grande of Tierra del Fuego (Argentina). Thanks both to environmental conditions favorable for colonization and an absence of effective control measures, today the estimated abundance is about 100 000 individuals. We have conducted molecular analysis of beavers from three areas of the Archipelago of Tierra del Fuego, in order to characterize the genetic structure of the invasive population. Our results from AMOVA (Fst= 0.104, p < 0.001) suggest that geographical barriers and large distances could limit gene flow among the populations. In the Tierra del Fuego National Park subpopulation, we found a positive and significant autocorrelation out to 800 m (r = 0.193, p = 0.003) and...

TAKAHASI, Shoko; MIYAHARA, Kazuro; ISHIKAWA, Hirosi; ISHIGURO, Naotaka; SUZUKI, Mitsuyoshi; 宮原, 和郎; 鈴木, 三義.
To estimate the maternal effects of dog breeds using mitochondrial DNA(mtDNA) haplotypes in the dogs with several clinical disorders, 600 base pairs of mtDNA D-loop region were amplified from 365 dogs and were determined for mtDNA sequences. The diversity of the 600-bp sequences was classified into 64 haplotypes, including 46 newly discovered haplotypes, and the haplotypes were grouped into four clusters I to IV. Lineage analysis using the mtDNA haplotype indicated that each dog breed genetically comprises one or a few mtDNA haplotypes. When the relationship between genetic background and occurrences of clinical diseases was estimated, canine lineage analysis using mtDNA haplotype revealed that the disorders distributed in the dominant mtDNA haplotypes of...

Rodrigues,Rosa; Schneider,Horacio; Santos,Simôni; Vallinoto,Marcelo; Sain-Paul,Ulrich; Sampaio,Iracilda.
The acoupa weakfish (Cynoscion acoupa - Sciaenidae) is a marine species of croaker with estuarine-dependent behavior, found in the western Atlantic from Panama to Argentina. It is one of the most exploited food fish on the northern coast of Brazil. In this study, DNA sequences were determined from the entire control region (D-loop) of the mitochondrial genome of 297 individuals collected during seven different months between December 2003 and August 2005 on the northern coast of Brazil (Amapá and Pará). Genetic variability expressed by haplotype (h = 0,892) and nucleotide (pi = 0,003) diversities were low compared to other heavily exploited marine fish species from the western Atlantic and eastern Asia. AMOVA depicted a lack of genetic structuring among...

Mitochondrial control region haplotypes of the South American sea lion Otaria flavescens (Shaw, 1800) BJMBR
Artico,L.O.; Bianchini,A.; Grubel,K.S.; Monteiro,D.S.; Estima,S.C.; de Oliveira,L.R.; Bonatto,S.L.; Marins,L.F..
The South American sea lion, Otaria flavescens, is widely distributed along the Pacific and Atlantic coasts of South America. However, along the Brazilian coast, there are only two nonbreeding sites for the species (Refúgio de Vida Silvestre da Ilha dos Lobos and Refúgio de Vida Silvestre do Molhe Leste da Barra do Rio Grande), both in Southern Brazil. In this region, the species is continuously under the effect of anthropic activities, mainly those related to environmental contamination with organic and inorganic chemicals and fishery interactions. This paper reports, for the first time, the genetic diversity of O. flavescens found along the Southern Brazilian coast. A 287-bp fragment of the mitochondrial DNA control region (D-loop) was analyzed. Seven...

Mitochondrial DNA D-loop sequence variation among 5 maternal lines of the Zemaitukai horse breed Genet. Mol. Biol.
Cothran,E. Gus; Juras,Rytis; Macijauskiene,Vale.
Genetic variation in Zemaitukai horses was investigated using mitochondrial DNA (mtDNA) sequencing. The study was performed on 421 bp of the mitochondrial DNA control region, which is known to be more variable than other sections of the mitochondrial genome. Samples from each of the remaining maternal family lines of Zemaitukai horses and three random samples for other Lithuanian (Lithuanian Heavy Draught, Zemaitukai large type) and ten European horse breeds were sequenced. Five distinct haplotypes were obtained for the five Zemaitukai maternal families supporting the pedigree data. The minimal difference between two different sequence haplotypes was 6 and the maximal 11 nucleotides in Zemaitukai horse breed. A total of 20 nucleotide differences compared...

Mitochondrial DNA variation in wild populations of Leporinus elongatus from the Paraná River basin Genet. Mol. Biol.
Martins,Cesar; Wasko,Adriane P.; Oliveira,Claudio; Foresti,Fausto.
Leporinus elongatus, a fish species widely distributed throughout the Paraná River basin in South America, is an important fishery resource and a valuable species in aquaculture programs. Despite its great economic importance, several wild populations have been suffering a drastic reduction. The comprehension of its population structure represents an important step for the conservation of these organisms in natural environments, and also for the selection of wild stocks to be used in hatchery programs. In order to understand the genetic-population structure of L. elongatus, D-loop mitochondrial DNA analyses were applied in six wild populations of the species. The results were used to estimate the levels of within and among population genetic variability....

Phylogeny of the Serrasalmidae (Characiformes) based on mitochondrial DNA sequences Genet. Mol. Biol.
Ortí,Guillermo; Sivasundar,Arjun; Dietz,Kelly; Jégu,Michel.
Previous studies based on DNA sequences of mitochondrial (mt) rRNA genes showed three main groups within the subfamily Serrasalminae: (1) a "pacu" clade of herbivores (Colossoma, Mylossoma, Piaractus); (2) the "Myleus" clade (Myleus, Mylesinus, Tometes, Ossubtus); and (3) the "piranha" clade (Serrasalmus, Pygocentrus, Pygopristis, Pristobrycon, Catoprion, Metynnis). The genus Acnodon was placed as the sister taxon of clade (2+3). However, poor resolution within each clade was obtained due to low levels of variation among rRNA gene sequences. Complete sequences of the hypervariable mtDNA control region for a total of 45 taxa, and additional sequences of 12S and 16S rRNA from a total of 74 taxa representing all genera in the family are now presented to...

Populations analysis of the Brazilian Sharpnose Shark Rhizoprionodon lalandii (Chondrichthyes: Carcharhinidae) on the São Paulo coast, Southern Brazil: inferences from mt DNA sequences Neotropical Ichthyology
Mendonça,Fernando Fernandes; Oliveira,Claudio; Gadig,Otto Bismarck Fazzano; Foresti,Fausto.
Sharks of the genus Rhizoprionodon can be considered some of the most important predators along the trophic coastal marine ecosystems and represent an important economic resource for the small-scale fisheries, especially on the Brazilian coastline. In order to analyze the population structure of the shark Rhizoprionodon lalandii of São Paulo, Southeastern coast of Brazil, levels of genetic diversity were identified by nucleotide sequence analyses of the mitochondrial DNA control region. The results obtained from this study present moderate values of haplotype diversity and low nucleotide diversity. Although the AMOVA tests (ΦST = 0.08394, P < 0.01) had shown slightly differences among the studied samples, evidence for the occurrence of population...

Tocantins river as an effective barrier to gene flow in Saguinus niger populations Genet. Mol. Biol.
Vallinoto,Marcelo; Araripe,Juliana; Rego,Péricles S. do; Tagliaro,Claudia H.; Sampaio,Iracilda; Schneider,Horacio.
The Saguinus represent the basal genus of the Callitrichinae subfamily. Traditionally this genus is divided into three groups: Hairy, Mottled and Bare-face, however, molecular data failed to validate these groups as monophyletic units, as well as raised some subspecies to the species status. This is the case of the former subspecies Saguinus midas midas and S. midas niger, which are now considered as different species. In the present study, we sequenced a portion of the D-loop mtDNA region in populations from the East bank of the Xingu and from both banks of the Tocantins river, in order to test the effectiveness of large rivers as barriers to the gene flow in Saguinus. According to our results, the populations from the East and West banks of the Tocantins...
